Background

Abdominal and intrauterine adhesions are common postoperative problems that may lead to serious complications. Current adhesives are usually double-sided, with poor wet adhesion, non-degradability and single function, which limits their application in preventing postoperative adhesions. In view of this, the research group of Yang Jianmin & Shi Xian'ai from Fuzhou University prepared a biomimetic microstructured Janus bioadhesive with a wet bonding inner layer and an anti-adhesive outer layer by combining electrospinning and adhesive materials for the prevention of abdominal and intrauterine adhesions. The relevant research results were published in the journal Advanced Functional Materials under the title "Bioinspired Microstructured Janus Bioadhesive for the Prevention of Abdominal and Intrauterine Adhesions".

The main point of this paper

1. A biomimetic microstructured Janus bioadhesive with a wet adhesive inner layer and an anti-adhesive outer layer was prepared by combining electrospinning and adhesive materials, named OD/GM@PG.

2. The wet adhesive strength and interfacial toughness of the Janus bioadhesive prepared by capillary suction and catechol-based adsorption strategy reached 98 kPa and 325 J m2, respectively, which are much higher than those of ordinary fibrin glue and cyanoacrylate glue.

3. The electrospun outer layer acts as a physical barrier, which has the effects of anti-adhesion and friction reduction. The bioadhesive has high wet adhesive strength and interfacial toughness, and also exhibits biodegradable, hemostatic, antioxidant, anti-inflammatory and healing properties.

In vivo experimental results show that the bioadhesive effectively prevents postoperative abdominal and intrauterine adhesions.

How is OD/GM@PG Janus bioadhesive prepared?

1. OD/GM@PG Janus bioadhesive is prepared by combining electrospinning and bonding materials.

2. The inner layer of Janus bioadhesive is made of catechol-modified oxidized hyaluronic acid (OD) and methacrylated gelatin (GM), while the outer layer is made of polycaprolactone (PCL) and gelatin.

3. The two layers are tightly connected through covalent/non-covalent bonding and mechanical locking of entangled polymers.

4. The preparation process involves various chemical reactions and physical interactions to achieve the desired asymmetric wet adhesion and anti-adhesion properties

What properties and functions does Janus Bioadhesive have?

1.Asymmetric wet adhesion: The inner layer of Janus Bioadhesive has strong wet adhesion properties that promote wound healing, while the outer layer acts as a physical barrier to prevent postoperative adhesions.

2. Biodegradability: Janus Bioadhesive is biodegradable and does not require a secondary surgery to remove it.

3. Hemostatic properties: It helps stop bleeding by absorbing blood from the wound and sealing the bleeding wound.

4. Antioxidant effect: Janus Bioadhesive scavenges reactive oxygen free radicals (ROS).

5. Anti-inflammatory and pro-healing effects: It helps inhibit inflammatory responses and reduce fibrosis, thereby promoting wound healing.
