Melatonin, as an endogenous hormone, has attracted increasing attention for its potential application value in the prevention of radiation-induced mucositis. Such applications include preventing mucosal damage, the appearance of ulcers, and the loss of proliferating stem cells caused by radiation. The oral mucosa, as a protective barrier between the oral epithelial surface and the external environment, has an important impact on the local delivery of drugs to the oral cavity.
Researchers have used thermal inkjet printing technology to improve the deposition accuracy of drugs on polymer oral films. The application of this technology allows melatonin to be deposited more accurately on polyvinyl alcohol (PVA) films, which is of great significance for improving the efficiency and accuracy of drug delivery.
The release and penetration of melatonin into epithelial cancer cell lines (TR146 cells) through an ex vivo oral mucosa model were explored in the study. The study found that the release of melatonin was lower in the presence of saliva, which may be due to the fact that saliva initiates the formation of mucosa through mucin-mucin interactions.
Drug controlled release system: Electrospinning technology can construct a controlled release system for traditional Chinese medicine to achieve controlled release and improve bioavailability of traditional Chinese medicine. This is particularly important for the application of drugs such as melatonin, because they need to be released at a specific rate within a specific time to achieve optimal efficacy.
Drug release rate: By adjusting the electrospinning process parameters, such as flow rate, voltage, collection distance, etc., fibers of varying fineness can be prepared, thereby controlling the release rate of the drug. This is critical for the design of delivery systems for drugs such as melatonin, as it allows researchers to customize drug release profiles based on therapeutic needs.
Drug stability: Organic solvents used in the electrospinning process may cause drug degradation, so new electrospinning methods need to be developed to avoid the use of organic solvents to maintain drug stability. This is particularly important for light- and heat-sensitive drugs such as melatonin.
The results show that as the sprayed droplets dry, the deposited melatonin crystallizes on the surface of the PVA film, which is consistent with previous studies in sprayed drug solutions. In attenuated total reflection Fourier transform infrared spectroscopy studies, the solid state of the deposited melatonin was further confirmed to be crystalline. Furthermore, no new bonds were detected, indicating that there were no new chemical bonds and interactions between melatonin and polyvinyl alcohol.
